Somatic embryogenesis of Euphoria longana var. "HAEW"

Softwood single node cuttings were inoculated in different treatments. It was observed that callus was formed in T1 MS + 400 mg glutamine + 800 mg PVP + 1 ppm Ki + 2 ppm NAA + 6 percent sucrose and T2 MS + 400 mg glutamine + 800 mg PVP + 1 ppm Ki + 2 ppm IAA + 6 percent sucrose. The callus formed was observed to have embryogenic cultures. Some of the embryonic cultures germinated into cotyledons. However, the cotyledons germinated precautiously. After 4 months, the cotyledons produced roots. These are being maintained in modified MS with 15 percent coconut water. The embryogenic cultures were transferred to modified MS with different levels of Ki and 2,4-D in order to produce globular embryos. These are being maintained under dark condition
