Use of rooted tomato leaves for direction of tobacco mosaic virus in tomato

Accumulation of tobacco mosaic virus (TMV) in whole plants and rooted leaves of TMV-resistant and susceptible varieties has been studied. Such methods of TMV detection as quantitative enzyme-linked immunosorbent assay (ELISA), inoculation of tobacco leaves (cv. Xanthi necroticum) and drop agglutination were used. Accumulation of the virus in the form of infective particles has been detected in the leaves of resistant hybrids F1 Verlyoka and Carlson rooted either before or after mechanical inoculation, but not in whole plants. Ordinary TMV infection process has been detected in rooted leaves from plants of two susceptible cultivars infected by TMV through seed. The conclusion was made that in whole plants of resistant hybrids with mechanical inoculation and in plants of susceptible cultivars with seed infection an inhibited TMV infection process develops. In rooted leaves this process becomes an ordinary one with accumulation of the virus in the form of infective particles
