Germination performance and loss of late-embryogenesis-abundant (LEA) proteins during muskmelon seed priming

Priming seeds in osmotic solutions has been used to improve the rate and uniformity of seed emergence. The objective of this work was to study the combined effects of oxygen supply, water potential and duration of muskmelon seed priming on subsequent seed performance. Muskmelon seeds (cv. Top Mark) were primed by immersion in aerated polyethylene glycol (PEG) 8000 solutions at different oxygen concentrations and water potentials for up to 10 days in stirred 500 ml vessels. After priming seeds were rinsed and dried back to original water content. Priming in water under high oxygen (= 50 %) decreased subsequent ability to germinate from 92 % to 20 % and increased t50 from 112 to 130 h.
