Distribution of anthocyanin in epidermal cells of grape [Vitis] berries

The distribution of anthocyanin in the epidermal cells of grape berries was observed microscopically. In black and purple-black grapes; Campbell Early, Muscat Bailey A, Black Olympia, Fujiminori, Pione, Kyoho, which had higher anthocyanin content, intensely pigmented spherical bodies appeared in the vacuoles very frequently. The bodies reached larger than 10 mu-m in diameter. On the other hand, in red grapes, the vacuoles were pigmented wholly and only small spherical bodies were observed in Delaware and Beniizu, but not in Olympia and Aki Queen at all. On the vine, abscisic acid (ABA) treatment induced accumulation of anthocyanin of Gros Colman' berries, and enhanced the development of spherical bodies and increased the frequency of appearance, whereas it stimulated only the coloration of whole vacuole in Flame Muscat. In the berry section of Gros Colman incubated in vitro, although the anthocyanin content was markedly increased by adding 0.6M sucrose and 0.1 mM ABA, little pigmented spherical body was appeared.
