Development of some bioassay methods for the screening of natural products from plants

Trichoderma harzianum Rifai proved to be an effective test organism for antifungal bioassay. Axenic cultures exposed to commercial synthetic antimycotic medicine ketoconazole and clotrimazole demonstrated differential inhibition of growth based on the concentration of the medicine. It also demonstrated a more potent effect of coumarin over salicylic acid and hydroquinone as shown by the inhibition of its conidia formation. Crude plant extracts could also be tested for antifungal activity. Extracts of Gliricidia sepium, like those of the clinically proven Senna alata, showed antifungal activity, Phytochemical tests showed anthraquinones, coumarin, and tannins are the major components of the G. sepium leaf. These may be responsible for the biological activity of its extracts determined by the brine shrimp general bioassay and the antifungal assay with T. harzianum as test organism
