Micropropagation of the cucumber (Cucumis sativus L.) hybrids Brunex and Bambina

The aim of this work was to test different media for their efficiency for micropropagation of parthenocarpic cucumber (Cucumis sativus L.) hybrids. Seeds of the cucumber hybrids Brunex and Bambina germinated in vitro. Shoot bud and shoot explants of the derived seedlings were cultivated in vitro on Murashige and Skoog (MS) media supplied with different combinations of growth regulators. The growth regulators used in these experiments were, the auxin 1-napthalenacetic acid (NAA) in doses of 0.0, 0,1, 1.0 mg/l, the cytokinin 6-benzylaminopurine (BA) in doses of 0.0, 0.1, 0.5, 1.0, 2.0, 5.0, 10.0 mg/l and the gibberellin gibberellic acid (GA3) in doses of 0.5, 1.0, 5.0, 10.0 mg/l. For the hybrid Brunex the highest shoot propagation rate was achieved when the medium was supplemented with 0.5 mg/l BA and 10.0 mg/l GA3 and no auxin. Every shoot with six buds produced 48 new axillary shoots within four weeks. In the case of the hybrid Bambina the highest shoot propagation rate was achieved when the medium was supplemented with a higher amount of cytokinin (1.0 mg/l BA) and a lower amount of gibberellin (5.0 mg/l GA3) and no auxin. Every shoot with six buds produced 52 new axillary shoots within four weeks. Elongation and rooting of the new shoots were carried out within four weeks on MS medium free of growth regulators for both hybrid
