Triticum durum cultivars grown in Slovakia have relatively low protein. To increase protein in wheat grains, the Hiproly spring barley cultivar with grains with 2-4 collaterally positioned aleuronic layers was used, and the cultivar was hybridised with Soldur, Slovakia's most productive durum wheat cultivar. Regenerants obtained from unripe embryos were amphidiploidized with use of colchicine 0.2 % solution. Twelve regenerants were obtained with fertility of ears and that of flowers ranging from 1-80 %. Mean protein content in regenerant grains was intermediate between content values of parental components. Two to three irregular aleuronic layers positioned collaterally and in the region of the abdominal crack were characteristic of the regenerants with higher protein content.
