Bulblet production of Lilium spp. in vitro

Bulblet production of Lilium spp. in vitro has been studied. Totally 16 cultivars were used. They were Apeldoorn, Chinook, Connecticut King, Corina, Enchantment, Golden Melody, Harmony, Hinomoto, Medallion, Mont Blanc, Orchid Beauty, Prominence, Red Night, Rose Fire, Rosita and Stering Star, Bulbscales were divided into 1x1 cm pieces and cultured in Murashige and Skoog media (MS) added with 0.1 mg/litre NAA. Bulblet production occurred in all pieces of bulbscale. Fresh weight of bulblets produced in vitro on MS media with 60 g/litre sucrose was the highest. Multiplication of bulblets was by half- and quarter-division. There was no difference in number of bulblet produced either in light or dark. Increasing concentration of sucrose enhanced bulblet multiplication. Maximum number of bulblet produced was found on MS with 60 g/litre sucrose. Daminozide gave negative result on bulblet production. Certain cultivars produced less number of bulblets with increasing daminozide concentration. Factors affecting bulblet emergence has been studied. The percentage of bulblet emergence was increased when treated with 100 mg/litre gibberellic acid (GA3) for 10 minutes. There was no difference in increasing percentage of bulblet emergence when treated in 50-200 mg/litre benzyl adenine (BA) solution for 10 minutes. Bulblets treated at 10 deg C for 1 month before planting could increase emergence more than 80 percent within 30 days.
