An FTIR/ATR study of the enzymatic wool treatment

Enzymatic treatment of wool in optimum, previously determined, conditions of pH, concentration of enzyme and treatment time of 30, 60 and 90 min have been investigated using FTIR/ATR spectroscopy coupled with second-derivative spectral analysis. The changes of peak intensity of cysteic acid, Bunte salt (S-thiosulfate), cystine-S-monoxide and cystine-S-dioxide were established. The decrease of amide I absorbance corresponds to splitting of peptide bonds after enzymatic treatment of wool. Quantitative analysis of particular oxidized cystine residues was done using FTIR/ATR second order spectra. The content of cysteic acid was increased from two-fold to eight-fold depending on treatment conditions. The mechanism of enzymatic treatment of wool may proceed by two possible mechanisms such as the mechanism of nucleophilic substitution and mechanism of beta-elimination, respectively.
