Application of flow cytometry for the study of plant genomes

The purpose of the paper is to review the field of flow cytometric analysis of plant cell nuclei and chromosomes. In the first part, basic principles of flow cytometry are reviewed. In the second part, methods for the analysis of nuclear DNA content and the use of FCM for ploidy screening and for genome size estimation, is discussed. In addition to the methodology and its applications, the impact of possible pitfalls is described. The third part concentrates on the use of FCM for the analysis and sorting of plant chromosomes, and on the potential of this technology for gene isolation and mapping
