Genotype of kappa-casein and lactalbumin locus for Czech Red breed and hybrids of the Czech Red breed crossed with Czech Spotted breed was determined by the PCR and RFLP methods. For Czech Red breed, the frequency of kappa-casein allele A was 0.46, of allele B, 0.48, and of allele E, 0.058. Frequency of genotype AA was 0.192, of BB, 0.192, EE and AB 0.500, AE 0.039 and EB 0.077. Expected and determined genotype frequencies did not differ significantly. For hybrids of Czech Red Breed and Czech Spotted breed cattle, the frequency of Allele A was increased to 0.614; frequency of allele B was 0.386; allele E dwindled from the cattle population. Frequency of hybrid genotype AA was 0.409, AB 0.409, BB 0.192. Expected and determined frequencies did not differ significantly. It was determined that there was an increase of homozygosity with hybrids. Monomorphism of alpha-lactalbumin locus was confirmed in European cattle breeds. Enlargement of number of heads of Czech Red breed, which belongs to the endangered gene cattle reserve, is recommended. It is also recommended to adjust breeding requirements for special small cattle populations
