The secretory activity of cultured ovarian granulosa cells as indicator of biochemical activity of porcine growth hormone

The effects of pGH addition (1-10.000 ng ml** (-1)) on the secretion of progesterone, androstenedione, insulin-like growth factor-I (IGF-I), oxytocin, insulin-like growth factor binding protein-3 (IGFBP-3), prostaglandin F2-alpha (PGF) and cyclic adenosine monophosphate (cAMP) by cultured porcine ovarian follicles or granulosa cells were assessed. It was observed that pGH significantly (P less than O.05) increased progesterone, IGF-I, PGF and cAMP output, whilst it significantly decreased IGFBP-3 and oxytocin release by cultured granulosa cells. Moreover, GH significantly stimulated PGF, cAMP, and inhibited androstenedione and oxytocin secretion by cultured ovarian follicles. These observations demonstrated an involvement of GH in direct regulation of the porcine ovarian secretory activity, as well as the usefulness of ovarian cell cultures for the assessment of pGH bioactivity
