High molecular weight glutenin subunits synthesis dynamics during the wheat grain maturation (Triticum aestivum L.)

The synthesis dynamics of high molecular weight (HMW) glutenin sub-units was investigated by analysing wheat grains of three winter wheat cultivars (Triticum aestivum L.), namely the Blava, Sana and Vlada varieties, sampled during maturation. Recommended by the international organisaion ISTA, standard vertical discontinual electrophoresis in the polycrylamide gel in the presence of SDS, followed by desintometric detection, was performed. Based on an increasing amount of the HMW glutenin sub-units, it was confirmed that biosynthesis occurs from the beginning of milky ripeness to the full physiological maturity. The biosynthesis rate was highest for the Vlada variety and lowest for the Sana cultivar. The biosynthesis of individual HMW sub-units was not synchronized in time. The synthesis of HMW sub-unit 9 of the Blava cultivar, HMW sub-units 2 and 12 of the Sana cultivar, and HMW sub-units 1,5,7 and 10 of the Vlada cultivar started only at spike dry matter of 43.77% to 47.37% and that of HMW sub-unit 9 of the Vlada variety commenced at 68.84% dry matter (collection of samples starting at 35% spike dry matter). The biosynthesis of individual HMW sub-units depends on the cultivar only. It is genetically conditioned and not dependent on the way in which sub-units effect the baking qualities of the cultivar
