Pomegranate (Punica granatum L.) propagation by in vitro method of tissue culture. [A short version of master thesis]

The introduction of pomegranate in culture in vitro was carried out by the method of micropropagation. Apical and axillary vegetative buds were used as primary explants. Isolates were grown on MS medium whose hormone structure was varied in dependence on the stage of the assay. Best initiation of the cultures was achieved on the medium containing 1.0 mg/l BAP and 0.1 mg/l NAA. All explants were thereafter transferred on to the stock medium which contained 0.5 mg/l BAP and 0.1 mg/l NAA. At the stage of multiplication, twelve different concentrations of hormones were varied across two parallel assays. Best growth and multiplication were achieved on media containing relatively low concentration of NAA (0.1 mg/l) and higher concentration of BAP (0.5 mg/l). Shoots were easily rooted on media containing five different concentrations of IBA (0, 0.1, 0.5, 1.0 and 2.0 mg/l). When the assay was completed, the rooted plants were transferred on to the soil substrate in a greenhouse. After, four weeks they were entirely adapted to ex vitro conditions. The highest degree of adaptation was achieved by shoots rooted on the medium containing 0.1 mg/l IBA
