In vitro establishment of Vaccinium cylindraceum Sm.

A micropropagation technique is developed and applied for the conservation of genetic variability within the endangered, natural populations of Vaccinium cylindraceum Sm. in the Azores. Only a solution with 0.3/ HgCl2 was effective to disinfect the explants from wild grown mother-plants; supplementing a fungicide solution (12/ benomyl) decreased contamination significantly. Explants collected in March were best suited for the production of axillary shoots. The medium Z2-80 produced the most constant and better results, regardless the population of origin of the explants. The first subculture should take place between the third and fourth week after initiation. Significantly increasing number of developing axillary buds was obtained with 7.38 microM 2iP in the initiation phase and 73.8 microM in the multiplication phase. 11.69/ of the shoots produced in the multiplication phase had juvenile leaves. Shoots were elongated in a hormone free medium and 71.43/ of the shoots rooted ex-vitro within 8 weeks.
