Utilization of different carbohydrates in sweet cherry (Prunus avium L.) micropropagation

Prunus avium L. cultivar 'Tommu' was in vitro preserved and micropropagated on Murashige-Skoog medium. The effect of carbohydrate source was examined, sucrose 30 g/l was compared to glucose 10, 20, 30, 40 and 50 g/l concentrations. Glucose 40 and 50 g/l provided better shoot proliferation than sucrose 30 g/l. Glucosecontaining media of 30-50 g/l were superior to sucrose-containing medium in shoot elongation. Percentage of shoots on different length levels was also affected by carbohydrates. At glucose concentration 10 g/l, most of the shoots were shorter than 10 mm while at 30, 40 and 50 g/l glucose about 80 per cent of shoots were in the length of 11...30 mm. In sucrose treatment the number of shoots less than 10 mm long was relatively higher than on glucose 30 g/l. The glucose-treated plantlets were more fragile and less lignified than sucrose-treated ones. As lignification is very important in microplants' acclimatization in greenhouse, glucose is not recommended for the last multiplication subculture before rooting phase
