Influence of medium content and in vitro preservation conditions on the productivity of potato genebank meristemplants

The regeneration of plants from microcuttings and the quality were more influenced by the content of medium as by preservation condition. The influence of different medium components were studied. The development of plants in vitro was more influenced by presence of kinetin as by increased concentration of potassium. Using the medium with kinetin 2.0...0.3 mg/l and sugar 25...30 g/l at preservation conditions 3...4 deg C, the multiplication cycle prolonged up to 100 days. The apperance of plants in vitro and inplastic rolls was influenced by medium content and by length of preservation period. The productivity of plants was not significantly influenced by medium content and by preservation conditions. The length of multiplication cycle could be exceeded by changing medium content and by considering specifity of genotype. There were not obtained deviations on morphological and economical characteristics of genotype. By the present research it can conclude that the medium-term preservation of potato genetic resources as meristemplants in vitro does not influence the genetic stability of genetypes
