Effect of culture medium to the growth and pathogeny of Metarhizium anisopliae (Metch.) Sorokin on Valanga nigricornis (Burm.) (Orthoptera : Acrididae)

An experiment was conducted at the Laboratory of Plant Pests and Diseases Department, Faculty of Agriculture, Islamic University of North Sumatra in 1996 to find the most suitable growth medium for and the pathogenicity of M. anisopliae to V. nigricornis. It used the CRD with six treatments, i.e. PDA, PDA + V. nigricornis extract, PDA + O. rhinoceros extract, corn, rice grain and rice meal and it was replicated four times. Results showed that the best medium for fungal growth was corn with growth area of 60.25 cm2 and conidia density of 8.42 x 10 at the power of 7 conidia/ml. V. nigricornis mortality was the highest, i.e. 85 percent and it occured 5.25 days after inoculation, the first among the treatments. Corn medium was also the most suitable for M. anisopliae mass culture, in small and big scale as well. Growth areas in rice meal, PDA + V. nigricornis extract, PDA + O. rhinoceros extract, PDA and rice grain media were 44.35, 35.92, 26.10, 21.79 and 18.25 cm2, respectively. The conidia densities were 7.24, 6.25, 6.11, 5.97 and 5.73 x 10 at the power of 7 conidia/ml respectively. V. nigricornis mortalities were 77.50, 67.50, 65.00, 57.50 and 55.00 percent, respectively, a while the first mortalities occured 5.75, 6.50, 6.75, 7.25 and 8.00 days after inoculation respectively

Suatu penelitian telah dilakukan di laboratorium Program Studi Hama dan Penyakit Tumbuhan Fakultas Pertanian UISU pada tahun 1996 untuk mengetahui media biakan yang paling sesuai bagi pertumbuhan jamur M. anisopliae dan daya patogenisitasnya terhadap hama V. nigricornis. Penelitian disusun dengan Rancangan Acak Lengkap (RAL) dengan enam perlakuan media biakan, yaitu PDA, PDA + ekstrak V. nigricornis, PDA + ekstrak O. rhinoceros, jagung, beras dan dedak padi yang diulang empat kali. Hasil penelitian menunjukkan bahwa media terbaik untuk pertumbuhan jamur adalah media jagung dengan luas pertumbuhan 60,25 cm2 dan jumlah konidia sebanyak 8,42 x 10 pangkat 7 konidia/ml. Jamur ini menimbulkan mortalitas V. nigricornis yang tertinggi, yaitu sebanyak 85 persen. Mortalitas pertama, yaitu 5,25 hsi juga disebabkan oleh M. anisopliae yang dibiakkan pada media jagung. Media jagung juga merupakan media yang paling sesuai untuk perbanyakan jamur, baik untuk skala kecil maupun secara massal. Pada media dedak padi, PDA + ekstrak V. nigricornis, PDA + ekstrak O. rhinoceros, PDA dan beras luas pertumbuhan jamur M. anisopliae masing-masing adalah 44,35, 35,92, 26,10, 21,79 dan 18,25 cm2. Jumlah konidia jamur yang dihasilkan adalah masing-masing 7,42, 6,25, 6,11, 5,97 dan 5,73 x 10 pangkat 7 konidia/ml.
