Bioluminescence assay in determination of total number of microorganisms in raw milk

Total number of microorganisms in raw milk was followed by means of the luminescent method by measuring of the light emission, proportional to the concentration of intracellular ATP. Having removed non-microbial ATP, the amount of ATP released from bacterial cells was indicated using the enzyme/co-enzyme complex, luciferase-luciferin, the addition of which initiated the luminescence. The results obtained were compared with those measured simultaneously using both the automatic method (Bactosan) and the standard plating method. For given type of luminometer, the ATP determination was optimized and interferences were eliminated. The differences in results of these three methods were statistically insignificant
