Selection of disease-free gladiolus produced by tissue culture

Studies on selection of disease-free gladiolus produced by tissue culture which was initiated from young flower stalks of gladiolus were cultured on Murashige & Skoog (MS) medium (1962) suplemented with Nitsch & Nitsch's organic addenda (1969) and 6-benzyladenine (BA) 2.0 mg/l. Callus were sampling and detecting ByMV by using immunoelectron microscope (IEM), Derrick & Decorate method. All callus which had already detected ByMV were cultured on the same medium to develope plantlets. Plantlets were detected again with IEM, Derrick & Decorate method. Cultures which were free from virus were multiplied to develope plantlets. Plantlets were transfered on MS medium suplemented with napthalene acetic acid (NAA) 0.1 mg/l to induce root. Plantlets were detected again with enzymed linked immunosorbent assay (ELISA). Short particles of ByMV were found in some callus but complete particles of ByMV were found in some plantlets. ByMV were not found in all plantlets which were multiplied from disease-free gladiolus. In vitro production of disease-free gladiolus var. Priscilla, Diemos and True Love on MS medium suplemented with Nitsch & Nitsch's organic addenda and BA 2.0 mg/l for 3 weeks, results from observation shown that gladiolus var. True Love had more weight, number and high of plantlets than the other two varieties.
