[Callus induction on garlic (Allium sativum L.) with indole acetic acid (IAA) and Kinetin]

The objective of this experiment was to evaluate the effectiveness and efficiency of in vitro callus formation on garlic explant using indole acetic acid (IAA) and kinetin KIN in Murashige and Skoog (MS) medium. Three levels of IAA (1, 2, and 3 mg/l) and three levels of KIN (1, 2, and 3 mg/l) were factorially arranged in a completely randomized design with three replicates. A piece of garlic vegetative bud with thickness of 1-2 mm was planted on MS medium in culture bottles. Callus culture was then maintained at temperature of 24-26 deg. C. under light intensity of 1000 lux with photoperiod of 12 hours of day/night. There was an interaction effect between IAA and KIN on success and rate of callus formations, and callus fresh weight. The effectiveness of callus production was obtained, subsequently on the combination of IAA 1 mg/l and KIN 1 mg/l, IAA 2 mg/l and KIN 1.5-1.75 mg/l, and IAA 3 mg/l and KIN 3 mg/l
