The authors examine the effects of culture media and sera on in vitro development of buffalo embryo. Cumulus-oocyte-complexes (COCs) were aspirated from ovaries of slaughtered Philippine carabao and utilized in this study. Results showed that mean number of COCs obtained per ovary was 3.1 wherein only 1.2 (36.6 percent) was classified as good quality COCs. No cleavages were observed in the media supplemented with locally processed buffalo calf serum. However, some zygote cleaved but stopped to develop up to 8 cells stages in mSOF supplemented with bovine calf serum. Embryos could develop up to the blastocyst stage in TCM-199 supplemented with bovine calf serum in vitro under the co-culture system with cumulus cells
