Genetic engineering approach to develop fungal resistance in rice

Genetic engineering is being conducted at PhilRice to improve the fungal resistance of high-yielding rice inbreds, new plant type elite lines and parentals for hybrids rice breeding. Chitinase and glucanase genes in binary vector pZ100 and the reporter gene green flourescent protein (gfp) have been successfully introduced into Agrobacterium tumefaciens EHA 105 through the freeze thaw and electroporation methods. Both methods are equally efficient in obtaining stable transformed clones as shown in the number of transformed colonies obtained and in the amount and quality of DNA recovered. The A. tumefaciens EHA 105 (pZ100) is presently being used to transform immature embryos and young inflorescence and calli derived from these explants. Preliminary results of the transformation experiments and their transient expressions will be reported
