Effect of sterilization method on infection level and regeneration in vitro of explants from bulbs of Frilillaria imperialis L

Explants from bulbs were grown on MS medium without any growth regulators. One step sterilization with Chloramine T at concentrations 2, 4 or 6 percent was compared with two-step procedure where application of chloramine was preceded by soaking bulbs in 0.2 percent Banlate solution. None of used Chloramine T concentrations increased significantly the number of sterile explants but soaking bulb pieces in 0.2 percent Benlate solution improved efficiency of sterilization, decreasing the number of fungal infections. However, use of Banlate at concentration 0.2 percent negatively affected regeneration ability of explants, decreasing propagation ratio and root formation. The highest number of sterile explants resulted from two-step sterilization where the use of 2 percent Chloramine T was preceded by soaking bulb pieces in 0.2 percent Banlate
