Effect of different auxins on rooting of apple dwarf rootstock M26

The aim of in vitro propagation is to produce healthy vigorous plants which are able to continue their growth ex vitro. Some species and cultivars survive in greenhouse conditions only if they have been rooted in vitro. In the experiments carried out in Eau Research Centre EVIKA we compared the effect of different auxins on rooting of apple dwarf rootstock M26 in vitro. Different concentrations (1,2,3 and 4mg/l) of naphtalene acetic acid (NAA) and indole-3-butyric acid (IBA) were compared. The best results were achieved with 3 mg/l IBA in the medium. Higher concentrations of IBA caused shoot tip necrosis, its lower concentrations reduced rooting percentage. Microcuttings grown on a medium containing NAA formed too long and brittle roots. NAA in higher concentrations resulted in low rooting percentage
