Study of the egg white lysozyme in sea turtle: Preparation and purification of green turtle [Chelonia mydas] lysozyme

The lysozyme in reptile is reported only for soft-shelled turtle egg white. To elucidate the distribution of lysozyme in reptile, we investigated the lysozyme in sea turtle (Loggerhead turtle, Green turtle, Hawksbill turtle) eggs. The electrophoretic analysis of egg whites revealed that egg whites of these turtles had protein bands around molecular weight of 14 kDa, but only green turtle had two protein bands at same position of C-type lysozyme. The lytic enzyme activity was found only in green turtle egg white. The lysozyme was purified from egg white of green turtle by extraction, pH treatment, and CM-Toyopearl ion exchange chromatography. Nine mg of lysozyme was obtained from 120 ml of egg white and the recovery of enzyme activity was 81.8%. The green turtle egg white contains lysozyme only 1% for total proteins in contrast to high ratio of lysozyme in soft-shelled turtle egg white that contains 32% of lysozyme
