Reference system for identifying bacteria simply based on available bacteriological properties

We developed a reference system to identify bacteria simply based on bacteriological properties. The system's database includes properties of bacterial species in from Bergey's Manual of Systematic Bacteriology, Bergey's Manual of Determinative Bacateriology, Guide to Plant Pathogenic Bacteria, and data that we have accumulated. For this study, we selected 94 types of bacteriological properties such as Gram reaction, the oxidation fermentation test, nitrate reduction, esculin hydrolysis, gelatin liquefaction, and the use of glucose, sucrose, and tartrate. The database currently consists of 90 plant pathogenic species and their pathovars, and 30 non-plant pathogenic species. Similarities between the test isolate and species in the database are compared by simple matching coefficients and their modified forms. Our study found the following: 1) To obtain results similar to those obtained by conventional identification, bacteriological properties used must number at least 50, including about 25 of culture-related and biochemical properties. 2) When the similarity index exceeds 90%, the isolate can be at least be related closely to a species in the database. 3) When the similarity index exceeds 95%, the species of the isolate can be identified. 4) Pseudomonas syringae pathovars cannot be effectively differentiated due to the similarity of their bacteriological properties
