High plant regeneration frequency obtained from scuttellum derived calli of 3 Thai rice varieties

Rice research program on genetic transformation and disease resistance has been recently set up in our department. In this congress, we present our first results on a method to enhance the regeneration frequency of 3 Thai rice varieties as Khao Dawk Mali 105 (KDML), Khao Ta Hang 17 (KTH), and Leuang Pra-tew 123 (LPT). Experiments followed a protocol of Sivamani et al., established for indica rice variety NTI regeneration using scutellum derived calli and based on modified N6 medium (Plant Cell Reports, 1996, 15:322-327). Regeneration frequency improvement for Thai rice varieties using the same initial material has been obtained by studying differents parameters such as cytokinine concentration in callus proliferation medium, abscisic acid (ABA) in maturation medium, and water stress in egeneration medium using a range of soild base concentrations (e.e. Phytagel). As for NTI, we found that the presence of cytokinine has enhanced significantly the regeneration frequency of at least 2 out of 3 varieties studied. Moreover, when the phytagel concentration was modified from 0.25 percent (w/v) to 0.6 percent (w/v) shoot regeneration frequency greatly increased in two rice varieties, KDML and LPT, but had little effect on KTH. With KTH, maturation of calli in a fresh medium containing 5 mg/l ABA for 7 days before transfer to regeneration medium was essential for the enhancement of shoot regeneration frequency. As a result of this study, these 3 Thai rice varieties have been regenerated at a high frequency (60 percent). Fertilily of the regenerated plant are under investigations and details of this study will be presented in the poster.
