Precondition for callus induction and in vitro organogenesis in water chestnut (Trapa japonica Flerov)

Callus induction and subsequent organogenesis were established in starch producing aquatic plant, water chestnut (Trapa japonica Flerov). Phenolic exudates released from the cultured explants of callus induction stage adversely affected the culture response. Phenolic accumulation in cotyledonary node derived callus cultured in different strength of MS medium containing different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) alone or in combination with benzyladenine (BA) was controlled by the addition of 2.0 mg/l phloroglucinol (PG) to the callus induction medium. Ascorbic acid (AA) was also less effective to reduce the phenolic accumulation than PG. Half-strength MS medium supplemented with 0.5 mg/l, 2,4-D, 20.0 mg/l casein hydrolysate and 2.0 mg/l PG induced the best callus. Plant organogenic was increased with vitamins (Biotin 0.05 mg/l and Folic acid 0.5 mg/l) onto the media containing half-strength MS salt and 0.05 mg/l BA under 28 and 14 hrs. photoperiod. Shoot multiplication has been achieved from the regenerated nodal shoot explants in the liquid media containing half-strength MS salt, 0.2 mg/l BA and 0.05 mg/l napthaleneacetic acid (NAA). Individual shoots were excised and cultured onto the rooting medium supplemented with 1.0 mg/l IBA and half-strength MS medium in liquid form. Rooted plantlets were acclimatized successfully in a water chestnut paddy field
