Assessment of genetic diversity and relatedness and identification of superior genotypes are the prime objectives of any crop improvement programme. Traditionally the assessment of the genetic composition of crop germplasm have been done on the basis of morphological characters which are time consuming and blurred by environmental influences. Recently, molecular markers such as RFLP, RAPD, AFLP, SSR, etc. have been used to assess the genetic diversity at the DNA level. Wild relatives of rice provide ample sources of resistance against abiotic and biotic stresses. Hence the present study is focused to assess the diversity and relatedness among the wild species of rice, using RAPD (Random Amplified Polymorphic DNA) and microsatellite markers (SSR). DNA was isolated from 40 genotypes and amplified through PCR (Polymerase Chain Reaction) using forty random primers (Operon Inc., USA) and twenty SSR primers (Rice Map Pairs from Research Genetics Inc., USA) following the standard protocols. The data on the banding pattern of both the RAPD and SSR analysis were scored and the data were analyzed using NTSY Spc.2.0 to construct a dendrogram following UPGMA method. The clustering pattern, the group constituents, the genetic diversity and relatedness among the genotypes and their implications in breeding programmes are discussed
