Development of inocula for treatment of wastewater from food manufacturing plants

Wastewater samples were collected from a food manufacturing plant. The samples were screened for indigenous microorganisms capable of degradation of organic matter. All bacteria isolated from the wastewater samples, together with existing bacterial cultures, were purified and then screened for growth on different wastewater concentrations and then for potential degrading ability towards carbohydrates, proteins and fats using artificial media. Out of the 42 isolates screened only 11 (V13, V14, M1, M2, M14, M15, M16, SL7, SL9, IN2, YW1) were found to be potential wastewater degraders. Chemical Oxygen Demand (COD) and organic matter content (carbohydrates, proteins and fats) were taken before and after 97-day incubation period (with shaking or aeration) for both lab-scale (250 ml) and scale-up (6 liters) set-ups. Among the 11 isolates chosen for actual treatment, only 2 (V13 and M16) were found to be the most efficient degraders. For the lab-scale experiments, isolate V13 was found to decrease the COD of pure wastewater by 65%, while M16 decreased the COD of pure wastewater by 75%. However, when inoculated on dilute concentrations of wastewater, both isolated, reduced the COD only by 7-36%. Isolate M16 reduced the COD of 1:3 (water to wastewater) wastewater concentration by about 36%, while only 21% reduced was achieved when wastewater was diluted 1:1. Isolate V13 isolated the COD of 1:3 wastewater concentration by about 20%, while only 25% reduction was achieved when wastewater was diluted 1:1. Increasing the volume of wastewater substrate to 6 liters generated an increase in the % COD reduction of both isolates. Isolate M16 reduced 1:3 wastewater concentration by about 93%, while only 76% reduced was achieved when wastewater was diluted 1:1. Isolate V13 reduced the COD of 1:3 wastewater concentration by about 90% whereas the 1:1 wastewater concentration was reduced by 82%
