Occurrence of apoptic bodies in preimplantation cow embryos

We analysed the early preimplantation normal and abnormal cow embryos (n=25) at the stage of morula, compact morula, early blastocyst, blastocyst and expanded blastocyst through the electron microscope to demonstrate the occurrence of apoptic bodies. To study the ultrastructure we fixed the embryos in 2.5 glutaraldehyde and 2.5 % paraformaldehyde, and postfixed them in 1 % osmium tetroxide, washed in cacodylate buffer, dehydrated by ascendant series of alcohols and embedded into Epon 812 resin. Ultrathin cuts were cut by ultramicrotome LKB - Nova and contrasted by the uranyl acetate and lead citrate. The electronogramms were made by the microscope JEM 100-CX II, Jeol at accelerating voltage 80 kV. We found out that the apoptic bodies in normal cow embryos occur intensively at the stages of blastocyst and higher, however, in abnormal embryos at the stage of morula already. The trophoblast of a normal blastocyst does not contain the apoptic bodies, however, they occur intensively in cells of the embryoblast. We found them also in cells of trophoblast and embryoblast in the abnormal blastocysts
