Heme-dependent catalase activity of lactobacilli isolated from Thai fermented food

Total of 33 strains of lactobacillus isolated from Thai fermented food were screened for heme-dependent and non-heme dependent catalase activity by oxygen bubble method. None of the strains tested has catalase activity in the absence of heme. 29 strains exhibited heme-dependent catalase and 4 strains were catalase negative. Of the 33 strains tested, only 3 strains exhibited high catalase activities by colorimetric assay, Lactobacillus sake SR 911, Lactobacillus plantarum P30-1, and Lactobacillus spp. F3-2. The catalase activity of aerobically grown cells showed maximum activity when added hematin at 30-50 mmoles per liter of growth medium. The kinetics of the catalase production studied in L.sake SR911 showed maximum activity at late stationary phase. This activity was pH independent in the range of 4.0-10 and was inhibited by azide. This catalase gene of L.sake SR911 was successfully cloned and sequenced and expressed in E.coli UM2 and is under studied its expression in local lactobacilli.
