Pre-screening of sugar beet varieties using molecular markers as an aid for DUS-testing

Choosing the right sugar beet variety is very important for both farmers and sugar industry. Their choice is based on the results of comparative testing of varieties for Value, Culture and Use (VCU) that is accomplished by specialised sugar beet institutes. Variety testing for Plant Breeders' Rights is based on morphological traits, which is labour intensive and expensive due the large trials (with at least 15 reference varieties) on different locations. Also, new candidate varieties are mostly near-hybrids and so more difficult to distinguish. The aim of this research is to develop a well defined set of molecular markers which allow a preliminary characterisation of new varieties during the winter before sowing, which can be an aid in the planning of the next trials and may also give a first indication for distinctness. With molecular markers it is possible to define the genetic identity of a plant in a quick and objective way. Three different molecular techniques form part of this pre-screening test: Amplified Fragment Length Polymorphisms (AFLP), Cleaved Amplified Polymorphic Site (CAPS) and Sequenced Tagged Microsatellite Sites (STMS). With 3 AFLP primer combinations, 13 CAPS markers and about 20 STMS markers a set of reference varieties will be characterised to develop a feasible pre-screening procedure.
