Effects of light intensity, nitrate and phosphate concentrations and pH on growth and carotenoid content of Dunaliella salina

Effects of light intensity, nitrate and phosphate contents and pH on growth and carotenoid content of Dunaliella salina were studied using D. salina clone number DS91008 which was isolated from salt ponds in Changwat Samut Songkhram, Thailand. The cultivation of D. salina in 20 percent NaCl-J/1 medium at more than 10,000 lux light intensity gave high carotenoid content comprising mostly beta-carotene with cell color changed from green to orange. Decrease in KNO3 content in the culture medium to 10 percent decreased growth but could accumulate carotenoid up to 137.2 pg/cell or 12 percent beta-carotene (dry weight). Change in KH2PO4 concentration affected growth but not carotenoid content. Optimal pH for algal growth was 7.4 while carotenoid content of D. salina was not affected by pH.
