Comparative study of extraction methods for leaf chlorophyll determination

Comparative study of extraction methods for leaf chlorophyll determination was investigated using eight sample plants. Completely randomized design with 8 replications was used. The total chlorophyll content from the leaf tissues of each sample plant was extracted by Dimethyl sulfoxide (DMSO) at 65 deg C, as described by Hiscox and Israelstam (1979), compared with the extraction by N, N-Dimethyl formamide (DMF) at room temperature, as described by Moran and Porath (1980). It was found that the total chlorophyll content from the leaf tissues of each sample plant which were extracted by both methods were not significantly different. DMSO extraction method, howerver, consumed shorter time than the DMF extraction method for all of the sample plants used in this experiment.
