The effect of quick freezing on development of mouse embryos at preimplantation stages

Английский. The aim of this experiment was to study the effect of quick freezing on in vitro development of mouse embryos at preimplantation stages. Embryos morulae stage and blastocyst were frozen in cryoprotectant 3.0 M ethylene glycol (EG) or 3.0 M glycerol (GLI) supplemented with 0.25 M sucrose in M16 medium with 4 mg/ml Bovine Serum Albumin (BSA). Both cryoprotectant had no effect on the in vitro development of mouse embryos at morulae stage and blastocyst to hatched blastocysts in control groups. The development of embryos after quick freezing using EG were better than that using GLI both in morulae embryos (65.38; 58.33 percent) and blastocysts (56.82; 49.02 percent). Показать больше [+] Меньше [-]индонезийский. Penelitian ini dilakukan untuk mengetahui pengaruh proses pembekuan cepat terhadap perkembangan embrio mencit tahap praimplantasi secara in vitro. Embrio tahap morula atau blastokista 0,25 M sukrosa di dalam media M16 yang mengandung 4 mg/ml Bovine Serum Albumin (BSA). Hasil penelitian menunjukkan bahwa perlakuan kedua krioprotektan tersebut tidak mempengaruhi perkembangan embrio. Hal tersebut terlihat dari berkembangnya embrio mencapai tahap hatched blastokista secara in vitro pada kelompok kontrol. Perkembangan embrio mencit setelah dibekukan di dalam EG menunjukkan hasil yang lebih baik dibandingkan dengan GLI, baik embrio tahap morula (65,38, 58,33 persen) maupun blastokista (56,82; 49,02 persen). Proses pembekuan cepat dapat dilakukan terhadap embrio mencit tahap morula maupun blastokista
