Functional studies of JEMMA and EMSY proteins

JEMMA is purative oncogene recently discovered as an interactor of EMSY protein, that negatively regulates the breast cancer BRCA2 protein. In the present study, we attempt to amplify the full-length JEMMA and EMSY in order to study their cellular functions. Four different constructs of JEMMA and EMSY have been generated and used in yeast two-hybrid screens to identify interacting proteins. These screens resulted in the isolation of a series of 9 proteins that interact with JEMMA and 7 proteins that interact with ENT domain of EMSY. Primary analysis of these interactors showed that most of them are involved in apoptosis. Two EMSY clones were generated, one with and one without exon 5. Overexpression of full-length EMSY (with exon 5) has a negative effect on cell growth compared to the empty vector. This negative growth effect is reduced when exon 5 is deleted.
