Tissue culture studies on japonica x indica crosses in rice

Because of the lack of cold tolerance, high-yielding indica rice varieties cannot be cultivated effectively during the extreme cold season. Three japonica rice varieties (Vailone Nano, Stagaree E 45, and Jimbu 4) were crossed as female parents with four indica varieties (Culture 1, TTB 14-1, Rongadoria, and IR64) as male parents. These crosses showed heavy shattering of immature seeds. Hence, 8-10-d-old immature embryos were cultured on MS medium supplemented with 0.25 mg L¯¹ 2,4-D. Both callusing and direct plant regeneration were observed after 4-5 d and 6-10 d, respectively, in most of the crosses. However, Jimbu 4 x Rongadoria showed the highest regeneration. Nodal segments from in vitro-raised tillers were cultured on MS + 0.5 mg L¯¹ BAP to produce more plantlets. In vitro-developed F1 plants exhibited intermediate characters of both parents. Several sets of ratoon tillers were also obtained from F1 plants, which showed early emergence of panicles. The segregating populations are being evaluated for cold tolerance under field conditions.
