Genetic analysis and field testing of elite rice cultivars transformed with the antisense waxy gene

The waxy gene is known to encode the granule-bound starch synthase (GBSS) responsible for the synthesis of amylose in rice endosperm. Several elite rice cultivars were transformed with the waxy gene in antisense configuration via Agrobacterium tumefaciens. Hundreds of transgenic lines were recovered in both japonica and indica rice. Analyses of polymerase chain reaction and Southern blot hybridization of the total DNA from transgenic plants confirmed that the transgenes had been integrated into the rice genome. The expression of the endogenous waxy gene was inhibited in some transgenic lines and amylose content decreased by different amounts. Genetic analysis suggested that the transgenes and amylose content in these transgenic lines could be inherited stably. Data from field testing showed that most of the transgenic lines had characteristics similar to those of the original cultivars. Several elite transgenic lines with low amylose content or no amylose (waxy type) have been selected for further testing and release.
