Fingerprinting of Indian scented rice by RAPD markers

The random amplified polymorphic DNA (RAPD) technique was used to characterize and assess the genetic relationships in 32 accessions of aromatic rice and three accessions of nonaromatic rice. All ten decamer primers used yielded scorable amplification patterns based on discernible bands. A dendrogram constructed using the Jacquard similarity coefficient and the UPGMA algorithm based on 176 reproducible polymorphic products ranging in size from 0.32 to 3.0 kb revealed a high level of variation among the accessions. Cluster analysis separated the 35 rice accessions into two main groups. All the aromatic rice clustered together with one exception. The similarity coefficient ranged from 0.370 to 0.811, showing a diverse gene pool. Accessions showing little morphological variation but having the same varietal name clustered together with one exception. Thus, RAPD markers can detect enough polymorphism, even among closely related aromatic genotypes.
