Isolation of chemopreventive agents against colon cancer from lemongrass

Total ethanolic extract of lemongrass was fractionated by solvent partition using hexane, ethyl acetate and butanol. Antimutagenicity of the 4 fractions (hexane-soluble, ethyl acetate-soluble, butanol-soluble and ethanol-soluble fractions) was evaluated. The hexane-soluble fraction appeared to be the most interesting because it had the strongest antimutagenicity against aflatoxin B1, IQ, Trp-P-1, Glu-P-1 and Glu-P-2 and was the most potent DT-diaphorase inducer. Oral administration of the hexane-soluble fraction prevented precancerous lesion formation in colon of male Wistar rats that received colon carcinogen (azoxymethane) during initiation and promotion stages. Rats on the hexane-soluble fraction diet were found significantly to have fewer azoxymethane-induced colonic aberrant crypt foci than those on azoxymethane alone. Interestingly, administration of the hexane-soluble fraction supplemented to the diet significantly decreased the number of aberrant crypt foci when given during both the initiation and promotion periods of azoxymethane-induced colon carcinogenesis. The results suggest that the hexane-soluble fraction affects the initiation phase of carcinogenesis as well as the growth and cell-differentiation.
