Susceptibility of poplar genotypes to Cryptodiaporthe populea (Sacc.) Butin and mycological traits of the fungus. [Master thesis]

The susceptibility of five poplar clones to the fungus Cryptodiaporthe populea (anamorph Dothichiza populea Sacc. et Br.) was studied from June 2001 to May 2003 at the experimental field. The investigations were carried out by the application of the artificial infections with mycelium of three selected isolates of the fungus in cortical tissue of the two-year-old plants of clones I-214, Pannonia, B-229, 182/81 and B-81 at the end of each month. The plants expressed a seasonal character of susceptibility to this pathogen. During the two-years of serial inoculations the highest percentage of the infections and the highest length of necrosis were during the period of vegetation rest (October-December), whereas the infections were not registered on plants which were inoculated during April, May and June. The research showed that no clones were immune on the onset of the fungus (fungus achieved infections in all clones, but in different degree). Separately, the same method of inoculation was applied on cutting of cl. I-214 on which the pathogenicity of nine isolates of Dothichiza populea in the conditions of greenhouse was tested in April 2002 and 2003. It was found that there were differences between isolates relating to the expressed pathogenicity. The appearance of pycnidia was not noticed on the artificially inoculated plants, but in the conditions of natural infection they were noticed on weak plants. The growth characteristics of isolates were studied by standard methods using solid semisynthetic and synthetic media in laboratory trials. The influence of the temperature on the fungus growth was studied by cultivation of fungus in polytermostat at temperatures between 5-30 deg C. The activity of the fungus in regard to the effects of the most important biogenic elements - carbon, nitrogen and sulphur were studied by the cultivation of fungus on solid synthetic medium.
