Relationship between plasma and faecal progesterone in gilts

Present study was performed to investigate the relationship between the levels of plasma and faecal progesterone (P sub(4)) metabolite during a normal oestrous cycle in gilts. Five crossbred gilts were used for the study. Feces and blood were collected every 3 days starting from the first day of standing oestrus (day 1) until day 19. P sub(4) metabolite in the feces was extracted using phosphate buffer solution. The measurement of P sub(4) levels in plasma and feces were carried out using solid-phase 125I-radioimmunoassay and its correlation was calculated by Spearmans correlation. It showed that the means faecal P sub(4) metabolite concentration on days 1, 4, 7, 10, 13, 16 and 19 of the oestrous cycle were 9.6, 17.9, 63.0, 255.6, 1006.6, 300.0 and 71.7 pmol/g, respectively. The levels of Psub4 in plasma collected at the same time as the feces were 1.9, 44.0, 78.5, 131.0, 88.5, 20.4 and 0.8 nmol/l, respectively. Variations in the levels of P sub(4) metabolite in the feces as well as P sub(4) in the plasma were relatively high among the gilts. On average, faecal P sub(4) metabolite was lowest on day 1 and highest on day 13 and plasma 14 was lowest on day 19 and highest on day 10 of oestrous cycle. The levels of plasma and faecal P sub(4) metabolite were significantly correlated (r=0.55, p LT 0.001). It was concluded that P sub(4) metabolite in the feces of gilts con be measured in place of plasma P sub(4), as it is significantly correlated to the level in plasma.
