[Biotechnological methods of rapid propagation of medicinal plants in tissue culture in vitro]

The purpose of the project consists in studying of features and laws of morphogenesis in vitro of big series of some perspective medicinal cultures and development of their new biotechnological methods of their effective microclonings and improvements. The purpose of the project defines following problems: 1) To receive aseptic cultures and to define optimum conditions of micropropagation (to estimate of morphogenetic activity of different types of explants, to define optimum conditions of rooting in conditions in vitro, structure of nutrient medium, types of growth regulators, physical conditions of cultivation for various species. 2) To develop methods of microclonal multiplication of studied rare medicinal plants. The objects of research were the following: (Agastache rugosa) – perennial grassy spicy-aromatic and medicinal plant. Stevia rebaudiana Bertoni - a perennial plant, wich is sweetener and herb. Melittis sarmatica KLOK. – perennial grassy medicinal plant brought in the Red book of Belarus. The following factors influencing on efficiency of process are studied: feature of a genotype of initial material, physiological age of initial explant, type and the size of explant, structure of a nutrient medium, hormonal balance of nutrient medium, role of biologically active substances not hormonal origin. The optimum mode of temperature and illumination is revealed. Methodical approaches are developed for each genotype in view of its specific features: 1) the scheme of preparation of vegetative material is developed for introduction in culture in vitro, 2) introduction in culture in vitro: development of conditions of sterilization of a material, 3) selection of cultivation environment , selection of cultivation conditions, selection of concentration of hormones in cultivation environment 4) receiving of active regeneration of shoots in culture in vitro, 5) actually clonal propagation with induction of plural shooting, shoots elongation, 6) rooting of shoots, 7) plants adaptation to conditions ex vitro
