Molecular identification of entomopathogenic nematodes found in pristine chestnut growing areas on the foothills of Etna (Sicily) [Castanea sativa Mill.; Sicily]

Morphological and molecular analyses (PCR amplification and sequencing of the 18S-5S ITS rDNA region) were conducted for the identification of populations of entomopathogenic nematodes of the genus Steinernema collected from pristine chestnut growing areas on the foothills of Etna. The results of these analyses indicated that the populations studied belong to the species S. feltiae. Comparisons of the ITS rDNA sequences of the Etna S. feltiae populations with the ones of S. feltiae deposited at Genbank showed a number of nucleotide variations characteristic of each population. These nucleotide variations may suggest the occurrence of indigenous S. feltiae variants in the surveyed chestnut sites

Alcune popolazioni di nematodi entomopatogeni del genere Steinernema, prelevate dal suolo di castagneti del monte Etna, sono state identificate a livello di specie sia mediante osservazione diretta dei caratteri morfologici, sia mediante sequenziamento di frammenti di DNA amplificati. Quest'ultimo approccio permette di amplificare un locus usando primer designati su sequenze conservate di geni rDNA. I frammenti sequenziati hanno permesso di identificare la specie S. feltiae e contenevano un numero di variazioni nucleotidiche caratteristiche per ciascuna popolazione
