Immune response evaluation trough cytokines quantification in persistent infected BVDV bovines [Bovine Viral Diarrhoea Virus]

Английский. Bovine Viral Diarrhoea (BVD) is an infectious disease responsible of serious economic losses in cattle farms. Despite intensive study about BVD and its etiological agent, there is not any instrument to control immunosuppression and congenital infection due to bovine viral diarrhoea virus (BVDV) infection. This is due to scarce knowledge about the effect of BVDV persistent infection on the host immune system. In this study, we investigated the expression of interleukin-8 (IL-8) and interferon-gamma (IFN-gamma) in 56 lymphoid organs from 12 BVDV naturally infected (PI) bovines through the quantification of mRNA by real time PCR. Values obtained were compared with the values observed in 19 organs from 3 healthy animals and with viral amount found in each organs. The results show lower levels of IFN-gamma in PI animals compared to healthy animals, while no statistical differences were found for IL-8. This study permitted to analyse and to confirm the interference of BVDV on immunological systems in natural infected animals. Particularly, downregulation of IFN-gamma showed the effect of BVDV on Th1 response

Nonostante la malattia sia ben conosciuta e studiata da molti decenni, ancora non è possibile controllare efficacemente né l'effetto immunodepressivo, né l'infezione congenita conseguenti all'infezione da virus della diarrea virale bovina (BVDV). Questa situazione è prevalentemente dovuta alle carenti conoscenze che si hanno sull'effetto dell'infezione virale persistente sul sistema immunitario dell'ospite. L'obiettivo della nostra indagine è stato la valutazione dello stato di immunodepressione in soggetti persistentemente infetti (PI) con infezione naturale da BVDV. Tramite real time PCR è stata valutata l'espressione genica dell'interleuchina 8 (IL-8) e dell'interferone gamma (IFN-gamma) in 56 organi linfoidi prelevati da 12 soggetti PI e comparata con quella di 19 organi prelevati da 3 soggetti sani. I livelli di queste due citochine sono stati poi correlati alla quantità di virus, quantificato sempre mediante PCR quantitativa. L'indagine ha mostrato valori di mRNA di IFN-gamma statisticamente inferiori nei campioni dei soggetti PI, mentre non sono state evidenziate differenze per IL-8. Ciò lascia ipotizzare una minore capacità reattiva della risposta immunitaria cellulo-mediata in animali PI con BVDV
