[Polypeptide composition of hard-to-extract protein fraction from chickpea seeds (Cicer arietinum L.) of different genotypes]

The polypeptide composition of hard-to-extract fraction of storage proteins from 31 chickpea genotypes (Cicer arietinum L.) of different geographical origin was studied with the help of electrophoretic separation in the gradient 6.5 12.7% polyacrylamide gel in the presence of SDS. The given fraction of storage proteins is presented by a small amount of peptides and consists of 8-23 components with low intensity of bands. Many of the revealed polypeptides are presented by the few numbers, and the components with the molecular mass higher than 150kD exist only in the footprint amounts. The genotypes were divided into 6 different groups according to their composition and similarity, and three genotypes (MDI 02401, MDI 02418 and MDI 02447) were individual. For the handling of the great volume of data, obtained from the electrophoretic separation of proteins, a mathematical program was used for determining molecular weight of the components, their intensity and the number of every polypeptide in spectrum. This program also alows to unify and automatically create the graphic picture of every spectrum on the electrophoregramme and to classify them according to their similarity and differences.
