Effect of different amino acids on the production of bacitracin by mutant strain of Bacillus licheniformis PCSIR-92

The development of microbial culture by mutation for industrial fermentation is widely used at present. The parent strain of Bacillus licheniformis PCSIR-88 produced 164 i.u./ml of antibiotic Bacitracin in shake flasks containing defatted soybean meal as substrate. After mutagenization with N-methyl-N-nitro- N-nitrosoguanidine, efficiency of the selected culture (PCSIR-92) for the biosynthesis of Bacitracin was greatly increased (420 i.u./ml). Effect of organic nitrogen on the Bacitracin formation by mutant strain PCSIR-92 was investigated in shake flasks. For this study nine different amino acids were supplemented singly or in combination with fermentation medium. Amongst all the amino acids tested, the antibiotic titre reached maximum 462 i.u./ml in the presence of 4 mM leucine containing isoleucine with strain PCSIR-92.
