Different effects of BAP and KiN on protein profiles of Morus alba leaf callus

Leaf discs (with or without mid-rib) of white mulberry (Morus alba) were subcultured in Murashige and Skoog media containing various concentrations of Kin, BAP, and NAA to study their influence on callus induction and organogenesis. The results obtained suggest that when same concentration of NAA was used with different source of cytokinins i.e. KiN instead of BAP, rooting was observed. In Kin and BAP treated callus, three different protein bands, were observed. The probable role of these proteins in root induction process is discussed. Further, when leaf discs with mid-rib was cultured in KiN and BAP containing media, development of adventitious root was observed from mid-rib in KiN treated discs. Root growth was more prominent in dark treatment than that of light. These results suggest metabolic regulation of two different cytokinins sources for the organogenesis.
